Choose the Right Reversed-Phase Column

Begin with the analyte’s hydrophobicity and the intended mobile-phase conditions. C18 is often selected as a practical starting point for general reversed-phase method development. When less retention is required, C8 or C4 may be appropriate. For methods involving aromatic compounds, isomers, or compounds requiring alternative selectivity, Phenyl, Biphenyl, Phenyl-Hexyl, PFP, or C18/PFP phases can be considered.
For highly aqueous mobile phases, select an aqueous-compatible C18 phase. For compatible biomolecule or macromolecule workflows, a large-pore option such as C18-BIO may be more suitable. C30 and PAHs phases should be considered where shape selectivity or dedicated PAH-related analysis is required.

Select the Appropriate Column Format

After choosing the stationary phase, select the column dimensions and particle size according to the analytical objective. Shorter columns may support faster methods, while longer columns can provide additional resolving power. Smaller particle sizes are commonly selected for higher-efficiency methods when the system and operating conditions are compatible.
