Effective method scale-up begins with a well-characterized analytical separation. Wherever possible, select a preparative or semi-preparative column with the same stationary-phase chemistry as the analytical method. This helps preserve relative retention and selectivity while allowing an increase in sample loading.
During scale-up, adjust the flow rate according to the larger internal diameter and maintain comparable linear velocity. Injection volume and mass loading should be increased gradually, because overloading can reduce resolution and lead to peak distortion. For complex mixtures, optimize loading capacity and fraction collection using the preparative system under the final mobile-phase conditions.
