Choose the Right Column for Your Method

Select the stationary phase based on the target analytes and the intended chromatographic mode. For conventional normal-phase separations using solvents like hexane or heptane, unbonded Silica, CN, Diol, or NH2 columns are appropriate choices. Diol and CN phases often provide faster equilibration and alternative selectivity compared with bare silica.
For highly polar, hydrophilic compounds, HILIC mode is generally preferred. Dedicated HILIC columns or HILIC-Amide phases provide strong retention for polar species using mobile phases with high organic content (typically acetonitrile) and a small proportion of aqueous buffer. NH2 columns are frequently selected for sugar and carbohydrate analysis in HILIC mode.

Select the Appropriate Column Format

Once the stationary phase is selected, choose the column dimensions and particle size that match the required resolution and system capabilities. Smaller particle sizes (such as 3 μm) and core-shell formats can provide higher efficiency for complex separations, while standard 5 μm particles offer robust performance for routine analysis. Internal diameter and column length should be matched to the sample load, flow rate, and analysis-time requirements.

Important Selection Note

Stationary phases such as CN and NH2 can often be used in either normal-phase or reversed-phase/HILIC modes; however, switching a single column between immiscible mobile-phase systems is generally not recommended.
